Water-induced hypo-osmotic test for the evaluation of canine sperm membrane integrity

چکیده
The purpose of this study was to investigate the efficiency of the water test to evaluate the functional membrane integrity of ejaculated canine sperm after collecting, cooling, and freezing-thawing. Twenty-five ejaculates were obtained from 12 stud dogs by digital manipulation. Sperm-rich fractions were evaluated, diluted, cooled at 5oC and frozen-thawed. A conventional hypo-osmotic swelling test (HOST) using a fructose solution (60 mOsm/l) was compared with a HOST using distilled water (0 mOsm/l), for evaluation of the functional integrity of the plasma membrane in fresh, cooled, and frozen-thawed semen. Distilled water detected a higher percentage of reacted sperm in fresh semen (91.2 ± 0.1%) than the conventional HOST (90.6 ± 0.1%; P < 0.05). For cooled and frozen-thawed semen, the water test was as efficient as the conventional HOST (81.2 ± 0.3% and 80.3 ± 0.3%, respectively), for detecting functional membrane integrity (P > 0.05). Regardless of the test used, there was a decrease in the mean percentage of reacted sperm after equilibrium and freezing-thawing (P < 0.05). In conclusion, this study demonstrated that water can efficiently replace conventional fructose-based hypo-osmotic media for evaluation of the functional integrity of the plasma membrane of ejaculated fresh, cooled, and frozen– thawed canine sperm.

P-77: HOST Patterns and Sperm Quality
Background: Following the expansion of human infertility, different procedures have been utilized for separation of normal sperm during ICSI (Intra cytoplasmic sperm injection). Hypo-osmotic swelling test (HOST) is one of the tests which have been introduced for assessment of integrity of sperm membrane when confronted with hypo osmotic condition. This test may also reflect intracellular organe...

Background: Intracytoplasmic sperm injection (ICSI) can be considered the solstice in vitro insemination technique because it has powerfully allowed the treatment of male factor infertility, but in this procedure, except visual morphological selection, there is no standardization for sperm selection. Recently, hypo osmotic swelling test (HOST) has been proposed to have the potential to select i...

The season may affect the values of fresh semen variables and therefore influence the success of cryopreservation. The aim of this study was to improve the evaluation of seasonal changes in semen quality in Spanish Black Castellana roosters maintained under natural environmental conditions. Semen was collected from 11 Black Castellana roosters (housed under natural photoperiod and temperature c...
متن کاملEffects of water- and lipid-soluble antioxidants on turkey sperm viability, membrane integrity, and motility during liquid storage.
Aerobic conditions are required to maintain the viability of turkey sperm in vitro. In mammalian sperm, excess oxygen during in vitro storage results in lipid peroxidation, causing membrane damage and reduced sperm motility and subsequent fertility. The effect of adding antioxidants to turkey sperm during liquid storage was studied. Semen was collected and pooled from 20 toms and antioxidants w...
